Avam® Reverse Transcriptase is specifically engineered to synthesize a specific complementary DNA strand from RNA or ssDNA.
The Avam® Reverse Transcriptase included in this kit is a modified, recombinant form derived from the Moloney Murine Leukemia Virus (M-MuLV), purified from Escherichia coli. This enzyme is specifically designed to synthesize a complementary DNA (cDNA) strand from either RNA or single-stranded DNA (ssDNA) templates in the presence of a primer. Notable features of the Avam® Reverse Transcriptase include its high thermal stability, making it suitable for use at higher temperatures (optimal activity at 50°C), and its ability to generate full-length cDNA strands, even from complex RNA templates. Additionally, it exhibits increased sensitivity in reverse transcription quantitative PCR (RT-qPCR) and reverse transcription PCR (RT-PCR) assays.
The enzyme is accompanied by a 10x RT Reaction Buffer containing Tris-HCl (pH 8.3), KCl, MgCl2, and DTT, providing optimal conditions for efficient cDNA synthesis.
Each unit of the Avam® Reverse Transcriptase is defined as the amount required to incorporate 1 nanomole of dTTP into an acid-insoluble material in 10 minutes at 37°C, using poly(A) x oligo(dT)12-18 as a template primer. In terms of performance, the enzyme exhibits high purity (>90%) and is free from DNase and RNase contamination, ensuring reliable results in downstream applications.
The Avam® Reverse Transcriptase is particularly beneficial for synthesizing full-length cDNA templates for RT-qPCR and two-step RT-PCR assays, as well as for cDNA synthesis in molecular cloning and cDNA library construction. It serves as a crucial tool in RNA analysis, enabling researchers
to study gene expression and RNA function with high accuracy and efficiency.
