The UniCycle™ RT-PCR kit is optimized and formulated for a single-step PCR reaction from an RNA template, enabling amplification without any additional steps. The kit is designed to merge the reverse transcription and PCR steps into a single process to generate exceedingly precise amplified DNA outputs saving your precious time.
The kit features a distinctive fusion of the revolutionary UniCycle™ reverse transcriptase, FusionTaq™ DNA Polymerase, UniCycle™ RT buffer and AVBlend™ allowing you to amplify low concentration and tricky GC-rich transcripts with zero to minimal optimization ensuring high yields.
The UniCycle™ enzyme mix features cutting-edge GenoScript™ RT enzyme and the FusionTaq™ DNA polymerase allowing a wide array of annealing temperatures, high affinity to RNA templates and a starting template concentration of as low as 1 picogram. The AVBlend™ is crafted to alter the melting patterns of nucleic acids thereby reducing the chances of secondary structure formation and RNase inhibition. The FusionTaq™ is designed to show zero activity at low and ambient temperatures and only activates after a hot start step during the thermocycling process which also inactivates RT enzymes.
The kit saves multiple setup, optimization and pipetting steps associated with PCR reactions and making a one-stop application for all molecular biology needs.
For 100 reactions:
UniCycle™ RT-PCR Enzyme Mix (1 X 0.2 mL)
5X UniCycle™ Buffer (1 X 1 mL)
dNTP Mix (1 X 0.2 mL at a concentration of 10 mM each)
Nuclease-Free Water (2 X 1.9 mL)
RNA template concentration range: Can reverse transcribe RNA transcripts from 1 picogram to 2 micrograms. FusionTaq™ Hot start: 95°C for 15 minutes. UniCycle Buffer: Contains K+ and NH4+ ions for enhanced yields. Reaction Steps: Prepare mastermix by adding all components to one tube, distribute and add template
The kit is recommended to be used for gene expression analysis, molecular diagnostic research, viral detection or single-cell RT PCR applications.
Typically a 50 µL reaction volume is recommended.
Yes, the unique characteristics of FusionTaq™ are activated when the PCR mix is hot-started at 95°C for 15 minutes.
The PrimeFactor™ additive present in the AVBlend™ increases the local concentration of primers around the template and also aids in stabilization of the primers post-annealing, therefore permitting FusionTaq™ to extend effectively compared to other Taq Polymerases on the market.
No, this kit does not feature a MasterMix but a one-shot MasterMix can be prepared by adding kit components and distributing them into different templates for highly reproducible results.
