Selectively isolate β-hemolytic Group A Streptococcus from throat cultures and upper respiratory specimens using InMicro® Prepared: Strep Selective II Agar (with colistin, oxolinic acid). Pacifico et al. demonstrated the isolation of β-hemolytic streptococci is improved with the use of a selective medium that inhibits oropharyngeal commensal microbial flora. In 1984, Pelts developed a selective medium containing colistin and oxolinic acid2. This medium was valuable for isolation of streptococci from mixed flora, resulting in increased isolation rates and early identification of isolates.
Clinical assessment alone is unreliable in distinguishing streptococcal and non-streptococcal pharyngitis3. Serious medical complications may follow pharyngitis caused by group A streptococci, making bacterial examination of the throat an indispensable diagnostic tool.
Use of Strep Selective II agar is recommended for selectively isolating β-hemolytic Group A Streptococcus from throat cultures and upper respiratory specimens.
This medium contains Tryptic Soy Agar supplemented with sheep blood as the base medium for luxuriant growth and enhanced hemolytic reactions of streptococci. Casein and soy peptones in TSA provide a nutritious source of nitrogen, amino acids, and peptides necessary for bacterial growth. Sheep blood includes growth factors and allows the detection of hemolytic reactions.
Colistin and oxolinic acid provide good growth from a very small inoculum and show no effect on streptococci while effectively inhibiting staphylococci, coryneform, and gram-negative organisms.
