Starch Agar was originally formulated by Vedder in the year 1915 for the cultivation of Neisseria. Heart Infusion Agar is used as the base for Starch Hydrolysis Agar with the addition of soluble starch. This medium is used for differentiation of Clostridium perfringens (positive) from other Clostridium spp.

Starch acts as the substrate which the organism may hydrolyze by producing amylase.
Starch Hydrolysis Agar contains beef heart infusion and casein peptone that supply nutrients necessary for the growth of all except the very fastidious bacteria. Yeast extract acts as a source of energy and provides additional nutritional requirements. Sodium chloride supplies essential electrolytes and maintains osmotic equilibrium. Starch is the fermentable substrate.
