Smith and Dawson added rose bengal to a medium with neutral pH for selective isolation of fungi from soil samples. Rose Bengal Agar with chloramphenicol is based on the formula developed by Jarvis for enumeration of yeasts and molds from food and water.

Contact I Rose Bengal Agar (with chloramphenicol) contains dextrose as the fermentable carbohydrate and a ready source of energy. Soy peptone supplies nitrogen and vitamin sources required for the growth of fungi. Chloramphenicol is added as a selective agent because of its heat stability and broad-spectrum antibacterial activity. Monopotassium phosphate is a buffer and magnesium sulfate provides sulfate and other trace elements. Rose bengal is an indicator and also acts as a selective agent which inhbits the growth of bacteria and limits the size of faster-growing molds. This helps to detect the slower-growing yeasts.
