Hajna and Perry developed EC Medium to detect Escherichia coli and other coliforms in water, milk, shellfish, and other materials. Feng and Hartman added 4-methylumbelliferyl-β-D-glucuronide (MUG) to conventional coliform assays. This improved detection rates of E. coli as fluorescence could be detected within 4-24 hours.
EC Medium with MUG is a liquid medium used to isolate and detect Escherechia coli. E. coli is an indicator organism that can indicate fecal contamination or unsanitary conditions.
EC Medium with MUG contains casein and meat peptones that supply amino acids, peptides and nitrogenous compounds required for growth of bacteria. Lactose acts as a ready source of energy and its fermentation leads to production of gas in the vial. Sodium chloride provides essential electrolytes and maintains osmotic equilibrium. Dipotassium phosphate and monopotassium phosphate are buffers that help control the pH. MUG is hydrolyzed by glucuronidase and yields a fluorescent end product, 4-methylumbelliferone.
