This two-enzyme system includes Uracil DNA Glycosylase (UDG) and Endonuclease VIII, each playing a distinct role in the process. UDG initiates the excision process by catalyzing the removal of uracil bases, resulting in the formation of an abasic site while preserving the integrity of the DNA backbone. Subsequently, Endonuclease VIII acts on the abasic site, cleaving the phosphodiester backbone both 3′ and 5′ to the uracil residue, thereby liberating the deoxyribose sugar.
