Product Description

Avam® DNA Ligase (E. coli) is a highly efficient enzyme used in molecular biology applications for joining DNA fragments at nicks and cohesive termini.

It catalyzes the formation of phosphodiester bonds between an adjacent 5′ phosphate and a 3′ hydroxyl of DNA ends, a crucial step in DNA repair and replication processes. This enzyme requires NAD+ as a cofactor and is particularly effective in sealing nicks and joining cohesive DNA ends.

The ligase is supplied in a buffer solution containing 10 mM Tris-HCl, 50 mM KCI, 1 mM DTT, 0.1 mM EDTA, and 50% glycerol, ensuring enzyme stability and activity.

Additionally, the accompanying 10Z DNA Ligase Reaction Buffer comprises 300 mM Tris-HCl, 40 mM MgCl2, 260 µM NAD, 10 mM DTT, and 0.5 mg/mL BSA at pH 8.0, providing optimal conditions for ligation reactions. Avam® DNA Ligase (E. coli) is characterized by its high specificity and efficiency, making it suitable for a wide range of applications in molecular biology research. It is commonly used in cDNA cloning, where it facilitates the joining of cDNA fragments during the cloning process.

Additionally, it finds utility in various DNA ligation reactions, such as the construction of recombinant DNA molecules, plasmid and genomic DNA cloning, and other genetic engineering techniques.

The enzyme’s robust performance, coupled with its broad temperature and pH tolerance, makes it an essential tool for DNA manipulation in the laboratory. Whether performing routine cloning procedures or more complex DNA assembly methods, Avam® DNA Ligase (E. coli) offers reliable and reproducible results, contributing to the success of molecular biology experiments.
