Overview

Isolate and enumerate Lactobacillus species and gram-positive cocci from clinical and non-clinical samples using InMicro® Dehydrated: M.R.S. Agar. In 1960, De Man, Rogosa and Sharpe modified a medium earlier formulated by Rogosa et al by eliminating tomato juice and created a medium which supported the growth of slower growing lactobacilli. Recently MRS Agar has been used in the clinical laboratory to differentiate certain strains of gram-positive cocci and lactobacilli from clinical samples.

‘Lactic acid bacteria’ are gram-positive, catalase and oxidase negative and are fastidious in their nutritional requirements. Growth is enhanced considerably by microaerobic conditions. Generally, the ‘lactic acid bacteria’ show delayed growth and smaller colony size than other microorganisms. They may be overgrown in non-selective media, especially if incubation is required for 2-4 days.

M.R.S. Agar is designed to encourage the growth of the ‘lactic acid bacteria’ which includes species of the following genera: Lactobacillus, Streptococcus, Pediococcus, and Leuconostoc. All these species can produce lactic acid in considerable amounts.

  • Can be used to differentiate certain strains of gram-positive cocci and lactobacilli.
  • Supports the growth of slower growing lactobacilli.
  • Low Selectivity: ammonium citrate and sodium acetate inhibit the growth of commensal microbial flora, gram-negative bacilli, and fungi, and improve the growth of lactobacilli.

This medium contains gelatin peptone and beef extract that supply essential nutrients and amino acids necessary for bacterial growth. Yeast extract is the growth enhancer and dextrose provide an energy source. Polysorbate 80 supplies fatty acids required for the metabolism of lactobacilli. Ammonium citrate and sodium acetate inhibit the growth of commensal microbial flora, gram-negative bacilli, and fungi, and improve the growth of lactobacilli.
