Selectively isolate and cultivate Mycoplasma species using InMicro® Prepared: PPLO Agar (with horse serum, amphotericin B, penicillin, and thallium acetate). Pleuropneumonia-like organisms (PPLO) were first studied and determined to be the causative agent of bovine pleuropneumonia. Edward and Freundt proposed the recent system of classification and nomenclature for PPLO, now referred to as Mycoplasmataceae. Kraybill confirmed that M. pneumoniae is resistant to methylene blue in agar medium while other species of Mycoplasma are inhibited.
The pleuropneumonia-like organisms (PPLO) are a group of pleomorphic, filterable, fastidious microorganisms which also lack a rigid cell wall. PPLO agar base was formulated to isolate and cultivate Mycoplasma species. The addition of yeast extract and unheated horse serum modified this formulation.
This medium contains beef extract infusion and peptone which supply nutrients that are necessary for growth of mycoplasmas. Sodium chloride maintains osmotic equilibrium. Yeast extract enhances the growth by providing B-complex. Normal horse serum provides a protein source.
