Hajna and Perry developed EC Broth to detect E. coli and other coliforms. They reported that this medium can be used to examine water, milk, shellfish, and other material from feces that pollute the environment. Previous methods were considered inadequate due to interference of injured cells that were sensitive to additives, temperatures and, selective media.
EC Broth is a liquid medium used to isolate Escherechia coli. E. coli is an indicator organism that can indicate fecal contamination or unsanitary conditions.
EC Broth contains casein and meat peptones that supply amino acids, peptides and nitrogenous compounds that are necessary for growth of bacteria. Lactose acts as a ready source of energy and its fermentation leads to production of gas in the vial. Sodium chloride provides essential electrolytes and maintains osmotic equilibrium. Dipotassium phosphate and monopotassium phosphate are buffers which help control the pH.
