Isolate and enumerate Lactobacillus species and gram-positive cocci from clinical and non-clinical samples using InMicro® Prepared: MRS Agar. In 1960, De Man, Rogosa and Sharpe modified a medium earlier formulated by Rogosa et al1 by eliminating tomato juice and created a medium which supported the growth of slower growing lactobacilli2. Recently MRS Agar has been used in the clinical laboratory to differentiate certain strains of gram-positive cocci and lactobacilli from clinical samples3-6.
‘Lactic acid bacteria’ are Gram-positive, catalase and oxidase negative and fastidious in their nutritional requirements. Growth is enhanced considerably by microaerobic conditions. Generally, the ‘lactic acid bacteria’ show delayed growth and smaller colony size than other microorganisms. They may be overgrown in non-selective media, especially if incubation is required for 2-4 days.
This medium contains gelatin peptone and beef extract that supply essential nutrients and amino acids necessary for bacterial growth. Yeast extract provides vitamin B complex and dextrose is the fermentable carbohydrate and energy source. Polysorbate 80 supplies fatty acids required for the metabolism of lactobacilli. Magnesium sulphate and manganese sulphate provide essential ions for multiplication of lactobacilli. Phosphates provide good buffering action in the media.
