Vera recommended using casein peptone in fermentation test media because of its high growth-promoting qualities. It was reported that peptone could be used with phenol red as the pH indicator in fermentation tests.
Phenol Red Broth is a complete basal medium prepared with phenol red as an indicator, which changes the medium to yellow when acid is produced. Carbohydrates may be added to the broth base in a final concentration of 1%. A Phenol Red Broth Base Control tube is used as a negative control for fermentation studies. A Durham tube may be inserted in a tube of Phenol Red Broth w/1% Sucrose to allow detection of gas production.
Phenol Red Broth w/1% Sucrose contains casein peptone that has high growth promoting qualities. Phenol red is the indicator which helps to determine the reaction by color change.
This medium also contains Sodium Chloride, Phenol Red, Demineralized Water and Sucrose. Sodium Chloride supplies essential electrolytes and maintains osmotic equilibrium of the medium.
