The Avam® Terminal Deoxynucleotidyl Transferase (TdT) provides the necessary components for efficient extension of DNA and RNA, useful for 3′ labeling and incorporation of dATP and dTTP with high efficiency. Avam® Terminal Deoxynucleotidyl Transferase (TdT) is a template-independent DNA polymerase used to add deoxynucleotides to the 3′ hydroxyl terminus of single or double-stranded DNA molecules. This pack includes 6000 U of TdT enzyme, 10X VividGreen™, and 2.5 mM Cobalt Chloride, facilitating the non-templated addition of dNTPs to 3′ termini of DNA. The presence of 1 mM Co2+ stimulates the tailing of the 3′-ends of DNA fragments. The enzyme efficiently extends blunt, 5′ overhanging, and single-stranded DNA and RNA. With its ability to incorporate dATP and dTTP with a fivefold higher efficiency than dCTP and dGTP, TdT is invaluable for various molecular biology applications requiring efficient DNA and RNA extension. It enables researchers to label 3′ ends with modified nucleotides, add homopolymeric tails to 3′-OH DNA ends, and label blunt ends of dsDNA breaks, as in the TUNEL assay.
