The Avam® Large DNA Construct Kit presents gravity-flow, anion-exchange columns designed for the purification of large-molecular-weight DNA with exceptional efficiency. Featuring an integrated ATP-dependent exonuclease digestion step, this kit ensures the selective removal of contaminating genomic DNA, enhancing the purity of the final DNA product. The purified DNA obtained through this kit is equivalent to that achieved by 2X CsCI gradient centrifugation and is ideal for transfection applications. The performance of the Avam® Large DNA Construct Kit is highlighted by its ability to yield DNA that is free from genomic DNA contamination, as depicted in the figure showcasing the efficient removal of genomic DNA. This purity enables accurate DNA quantitation, facilitating sensitive experiments conducted under defined and reproducible conditions. Utilizing an optimized gravity-flow procedure, the kit’s anion-exchange resin provides DNA purity comparable or superior to that obtained through CsCI gradient centrifugation. Prepacked Avam® AnioFlow tips operate via gravity flow, minimizing hands-on time and eliminating the need for toxic substances like phenol, chloroform, and CsCI, ensuring user and environmental safety. The procedure involves alkaline lysis of up to 500 mL culture, followed by an integrated digestion step with ATP-dependent exonuclease for selective removal of contaminants. Subsequently, the sample is loaded onto the anion-exchange tip, where plasmid DNA binds selectively. After impurities removal and elution in high-salt buffer, the DNA is concentrated and desalted, ensuring pure plasmid DNA suitable for various applications. The DNA purified using the Avam® Large DNA Construct Kit finds utility in a wide range of applications, including subcloning, shotgun library preparation, direct sequencing of large-construct DNA, and transfection, making it a versatile tool in molecular biology research.
